Neuropeptides are one of the largest families of extracellular messengers involved in various physiological mechanisms and pathological conditions. They have an extensive phylogenetic history. These small proteins are produced and released by neurons, exerting strong effects on adjacent glial cells and neurons (i.e., paracrine signaling). Additionally, neuropeptides can be released from neurons to gain access to the hypothalamic-pituitary-portal vessels (i.e., neuroendocrine factors) or enter the general circulation (i.e., endocrine hormones). They can act as hormones, neurotransmitters, and paracrine factors. Unlike classic low-molecular-weight neurotransmitters, neuropeptides are exclusively produced within the cell without local synthesis in nerve endings. In most instances, several different neuropeptides are encoded by a single continuous messenger RNA (mRNA), which is translated into one large precursor protein (polyprotein). Like other secretory proteins, neuropeptides or their precursors are processed in the endoplasmic reticulum before moving to the Golgi apparatus to be processed further. They leave the Golgi apparatus within secretory granules and are transported to terminals via fast axonal transport.
